Oh sure, I'm sorry I didn't write how the problem was solved (and yes,
it's kinda embarassing ;-)

the file containing the relay_domains had not the proper syntax to
make it a real hash file as I first assumed it was.

domain1 OK
domain2 OK
...

should be the style (as like virtual) but I had something like

domain1, domain2,...

and this was the reason why it didn't work. And honestly, I have read
most of the man pages and the sample files and online docu and I still
didn't know how to have the correct syntax. Perhaps a faq entry like
"how to correctly syntax your files for use with hash/pcre/btree/..."
would be helpful for other newcomers?
