No, at least the IMAP part reads maildir format.
The database needed is for authentication information. The advantage of
Courier over virtual-pop3d is that you can specify any path you like.
However, the disadvantage (if it is one for you) is that you have to
deliver to maildir, so you'd have to convert any existing mbox setups.
