This is described in sample-local.cf.gz:

# The mail_spool_directory parameter specifies the directory where
# UNIX-style mailboxes are kept. The default setting depends on the
# system type. Specify a name ending in / for maildir-style delivery.
#
#mail_spool_directory = /var/mail
#mail_spool_directory = /var/spool/mail

Place it in main.cf.
Note: this is a different parameter than home_mailbox.
