Yes and no. ;-)

It's easy with most PC based NIC's and drivers. Some OS's and features
actually require it, so it's built into most NIC chipsets and drivers.

Of course, once the frame has traversed a router, the original source
MAC address is gone, so it's usually irrelevant unless your target is
on the same subnet as you.
